An influence of mesenchymal stem cells transplantation upon cognitive function restoration after stroke in rats

Cell therapy is a promising modern attempt that can improve recovery after stroke. Mesenchymal stem cells are supposed to be an effective agent of the therapy.

 

Experiments were carried out on inbred Wistar-Kyoto rats. MSCs were isolated and expanded in culture. Animals were subjected to middle cerebral artery occlusion (MCAO), followed by injection of singeneic rat MSCs into tail vein 3 days after MCAO. Control group animals received PBS injection (negative control). A degree of cognitive functions alteration was studied by testing in Morris water maze. The test sets were carried out during 6 weeks after MCAO. MSC transplantation improved survival level; quantity of rats survived until 6 weeks after operation in cell therapy group was 1.7 times higher compared to control. The test results also improved. After MCAO animals didn't loose ability to learn: the time spent in looking for platform gradually decreased during the test session. Cell therapy group animals needed less time to find a platform comparing to control group. Moreover, experimental group rats worked out an optimal strategy for platform seeking by the 5th day of study and successfully used this strategy during the following testing. Control group animals showed worse results. Basing on these results we can suggest that MSC transplantation had an influence on cognitive function and led to preservation of learning ability.

 

We consider that the obtained results are connected with a phenomenon of decreased tissue defect in the experimental group, which was confirmed by a histological analysis.
